Description
General Description
Boc-NH-PEG-COOH is a versatile heterobifunctional PEG linker designed for orthogonal synthetic strategies. The terminal carboxylic acid group can be readily activated using carbodiimide chemistry (EDC/NHS) for coupling with primary amines on peptides, proteins, antibodies, polymers, nanoparticles, and biomaterial surfaces, forming stable amide bonds.
The opposite terminal amine is protected with a Boc group, preventing undesired side reactions during synthesis. After the desired conjugation is completed, the Boc group can be selectively removed under acidic conditions (typically trifluoroacetic acid or HCl in dioxane) to generate NH2-PEG-COOH, providing a free amine for subsequent modification.
The hydrophilic PEG spacer improves water solubility, molecular flexibility, and biocompatibility while reducing steric hindrance and minimizing nonspecific interactions. Boc-NH-PEG-COOH is widely employed in peptide chemistry, PEG linker synthesis, hydrogel preparation, nanoparticle functionalization, targeted drug delivery, and advanced biomaterials development.
